The EPR spectroscopic parameters of native fenugreek amine oxidase and its ?-nitrophenylhydrazine are consistent with the presence of Cu II ; tetragonal co-ordination with axial symmetry as published for other amine oxidases Dooley et al., 1991; Matsuzaki et al., 1995; Steinebach et al., 1996 ; . Estimation of the copper content of the native enzyme and the results of spectrophotometric titrations indicate the existence of one Cu II ; topa quinone site in each subunit of the dimeric enzyme. The c. 50% lower content of Cu II ; the enzyme 7-nitrophenylhydrazone is somewhat unexpected and could be attributed only to a release of Cu II ; from the active site of the derivatized enzyme. The observation of signals of Mn II ; ions in the EPR spectra of both enzyme and and its 7-nitrophenylhydrazone was, from the beginning, somewhat surprising and it was thought to be an impurity in the samples. However, the signal persisted even in enzyme samples that were repeatedly purified by FPLC using metal-free buffers. The explanation came with the recently published crystal structure of pea seedling amine oxidase which revealed the presence of a second metal site close to the protein surface which might be occupied by manganese Kumar et al., 1996 ; . This site is highly conserved among amine oxidases from various sources and it also exists in the E. coli amine oxidase where it is thought to be occupied by Ca II ; Parsons et al., 1995 ; . In the past, Mn II ; signals were found in the amine oxidase from human placenta, which was originally reported as a Cu metalloprotein Crabbe et al., 1976 ; . Considering the close sequence similarity of Fabaceae amine oxidases, the EPR data provide experimental evidence for the existence of another metal site in the fenugreek enzyme. From the results obtained by atomic absorption spectrometry, however, only about 10% of these sites is ocupied by Mn II ; the native enzyme. Remaining sites may be occupied by Ca II ; coli ; or other bivalent cationts. Therefore any role of manganese in the catalytic cycle of the amine oxidase is unlikely. Following from the above discussion, the recently found plant amine oxidase from fenugreek seedlings can be rapidly purified using the improved purification protocol.
