Cell lines were selected in a step-wise process for approximately one year and were maintained in 12 ng depsipeptide. For these analyses, cells were cultured in the absence of drug for two weeks. In cell lines selected in depsipeptide alone Dp cells ; , the observed IC50 was approximately 8 ng ml for cells, representing greater than 10-fold resistance to depsipeptide. These cells were sensitized to depsipeptide when treated with the Pgp inhibitor, valspodar Fig. 5A ; . The observed IC50 for the cells selected in the presence of verapamil DpVp cells ; exceeded 50 ng ml. Combination with valspodar did not significantly increase sensitivity to depsipeptide. Expression of Pgp was evaluated by using an antibody, MRK-16, that detects Pgp on the cell surface by flow cytometry and function of Pgp was evaluated by the efflux of rhodamine 123. No Pgp was detected in the parent cell line, consistent with the observation that valspodar did not increase sensitivity to depsipeptide by cytotoxicity assay. As expected, Dp cells selected for resistance to depsipeptide alone had significant levels of detectable Pgp and rhodamine 123 efflux. Cells selected for resistance to depsipeptide in the presence of verapamil had slight expression of Pgp and minimal rhodamine efflux, indicating that resistance is not mediated by Pgp. Increased histone acetylation was observed in both the Dp and DpVp cells following treatment with 25 and 100 ng ml depsipeptide, respectively, suggesting that in both cell lines, any reduction in histone acetylation that might result from drug resistance mechanisms can be overcome by increased doses of depsipeptide.

The blood-brain barrier represents a formidable barrier for drug delivery to the central nervous system. Tight junctions, the lack of fenestrations, and a low transcellular pinocytic index severely limit the accumulation of macromolecules in the brain Levin et al., 1980; Seymour, 1992 ; . Surprisingly, several groups have been able to show that even tumors located in the brain have a "leaky" microvasculature, although pore sizes are significantly smaller 100 380 nm ; than those seen with tumors located elsewhere in the body 200 780 nm; Siegal et al., 1995; Hobbs et al., 1998 ; . In a brain tumor model, Gabizon and coworkers were able to show high levels of SSL DOX accumulation in the tumor Siegal et al., 1995 ; . Fischer rats injected with SSL DOX at a dose of 6 mg kg showed maximal accumulation at 48 h DOX g of tumor tissue. This was 15-fold higher than the levels observed after the administration of an identical dose of free DOX 0.8 g g at which were not different from levels found in the normal brain contralateral hemisphere ; . There was no accumulation of SSL DOX in the contralateral hemisphere, and in brain tissue immediately adjacent to the tumor, levels were 2 g g tissue up to 70 after injection but gradually increased to a maximum of 4 g 120 h. These results suggest that even in the tightly regulated central nervous system, a high tumor vascular permeability can be exploited for carrier-mediated drug delivery. B. Rate and Extent of Accumulation in Tumors The rate and extent of drug accumulation in tumors vary depending on dose, formulation, and tumor type and vicodin.
